Most of the higher end sousaphones include upper tuning slides on all 3 valves, so you can do slide pulling. Both the first and third valve slide normally have both upper and lower tuning slide. I'd probably tune the first and second slides so that 1+2 is in tune, then tune the third valve so 2+3 is a tad flat. Another thing I'd do is be aware of what notes you need to play based on the key you are in. For instance if you are playing in the key of C you are not likely to run into a whole lot of F#'s or C#'s so you can then tune the third slide so that 1+3 is in tune, since you are going to see a lot of low C's in that key. On the other hand if you are playing in the key of Db then you want to tune the third slide so Db is in tune, and you probably won't need to play low C or low F a whole lot in that key.
