I think that coating it with layers of polyeurathane will defeat much of the purpose of playing on a wooden mouthpiece. Put enough poly over it and thats what your playing, poly.
You could for that matter, coat a regular mouthpiece with poly and play that. Better I think to choose a wood that doesn't need such protection. Perhaps a light coat to make it water resistant, but still leave the wood feel. Are you mating it to a metal shank? You could find someone who has a good wood lathe and copy a rim or rim and bowl and attach it to the shank off of another moutpiece.
