What was the reason behind the request of the conductor? Did he expect the other tubist to be there with a contrabass tuba?

You mention a march out of the Central European tradition. That tradition uses F as well as BBb tubas, but when only one tuba, then the BBb has the preference.

As a side comment: I have not the smallest shade of doubt, that the YFB 621 is a superb tuba within its intended areas of purpose. But while we often put F and Eb tubas into the same category of The Basstuba, I cannot avoid mentioning, that if I should cover an ever so small concert band as the single tubist playing a basstuba, then I would rather do it from an instrument like the Besson 981 Eb, even if it shares its .689 valve bore with the YFB 621. But of course I would rather use our common acquaintance, the YM BBb.
