On my 3302, I have the 5th tuned to a really flat whole step -- essentially a minor 3rd. Using 235, the low E is smack in tune, and the B natural above is quite good, too (although it clashes badly when in a section of 3- and 4-valve BBb tubas and their inherently sharp B fingerings).

One tip for getting those lowwww notes' fingerings is to play them up an octave, but with the same ham-fisted fingerings. I did this to find the combinations for my tubas, and playing down there suddenly became a lot easier (and more fun!) because the pitch centers were finally correct.
