I graduated from Eastman a little while back. I never got into All-State but got into Eastman. As has been said numerous times previously, play what you sound best on. I would suggest a piece that is challenging with contrasting styles as well as etudes by Bordogni or Blazevich.
To more accurately answer your questions I would suggest hanging out at the University of Wisconsin or University of Wisconsin-Milwaukee. There are some great students at those two schools close to where you live and it may give you a good idea of how well college tuba players play compared to where you are at right now.
Manhattan is generally a bit harder to get into than Mannes. Eastman's numbers for accepted tuba students has varied a lot from year to year over the course of the last ten years.
